PacBio amplicon sequencing was used as an orthogonal method to confirm the performance of Labcorp Pan-Heme NGS (LPH) panel to detect somatic variants in hematologic malignancies across 141 clinically relevant genes. 207 variants identified by LPH with an emphasis on low frequency variants were assessed by Pacbio with high concordance observed between LPH and PacBio. This study highlights PacBio’s utility as a cost-effective method for confirming variants in clinical diagnostics while demonstrating the high sensitivity and accuracy of LPH panel to detect variants ≥3% allele frequency.
